The objective of this study was to evaluate various polymers as a sustained release and mucoadhesive component of MCN mucoadhesive buccal tablets. The drug has poor aqueous solubility. The aim of this work was to developed and characterized a mucoadhesive controlled release buccal tablet of MCN. The buccal route was chosen because of its good availability, lustiness of the epithelium, facile removal of the dosage form, relatively low enzymatic activity, natural clearance mechanisms for elimination of the drug from buccal area, satisfactory patient acceptance and avoiding the hepatic first pass metabolism. Apart from the overall increased bioavailability because of bypassing the first pass effect and sufficient time to produce therapeutic effect, an important advantage of buccal delivery for MCN is potentially better control of plasma levels, typically low variation in bioavailability, reduced costs of the drug because of application of much lower doses than necessary for oral products. Among different trials with direct compression, trial F10 had satisfactory in-vitro dissolution profile in comparison to the innovator. So, these batches tablets were selected for the preparation of tablets. Stability study was carried out for the optimized formulation at 40° C / 75% RH for 3 months. The result shows no significant change in physical and chemical parameter of the tablet; hence the formulation was found to be stable.
