Abstract: A majority of drugs developed and used today suffer from poor bioavailability due to poor aqueous solubility or low dissolution rate. One of the main focus of pharmaceutical technology is developing novel strategies to enhance the solubility of poorly water soluble drugs. Nano suspensions are a promising strategy for improving the dissolution rate and oral bioavailability of poorly water soluble drugs by reducing the particle size or transforming drugs from crystalline to an amorphous state. In the present time an increasing attention has focused on nano suspensions considering these general advantage of nanotechnology ,the main objective of the present investigation was to prepare Nanosuspension of Oxcarbazepine which is widely used as antiepileptic drug belongs to BCS class II (low solubility, high permeability). Nanosuspension of Oxcarbazepine we are produced by combining nano precipitation and high pressure homogenization technique with using polymer like poloxamer 188,soluplus ,PLGA etc. All the suspension prepared with the different drug : polymer ratio showed a significant decrease in particle size with increase in the number of homogenization cycles and pressure. Nanosuspension were prepared and analyzed for particle size, zeta potential ,entrapment efficacy and in vitro drug release study. Batch (PLG2) drug : polymer:stabilizer (1:0.5:0.5) ,shows particle size (229.1nm) ,P.I.(0.411 ) , zeta potential (-0.2 ), % entrapment efficacy ( 73.86 ) and better drug release. Oxcarbazepine nanoparticles were produced by converting nanosuspension into dry powder by lyophilization technique. The physicochemical properties of drug and its nanoparticles were characterized by SEM,XRD,FT -IR,DSC, in - vitro drug dissolution and saturation solubility.The DSC and XRD results indicated that nano precipitation led to amorphization of oxcarbazepine. An increase in the stability of nanoparticles was also assured by sufficient adsorption of the polymer on to the surface.
